Singapore–Jakarta is about 1.5–2 hours with a 2–4 hour layover, then 3–3.5 hours to Ambon. Singapore–Makassar is 2.5–3 hours with a 1.5–3 hour layover, then about 1.5 hours to Ambon.

Layover length
A tight Makassar connection saves time on paper and costs a day if the first leg slips. If your dates are fixed, take the longer Jakarta layover.
The Ambon night
Neither routing gets you to Banda the same day. Budget USD 15–40 for a room and treat it as part of the fare.
Budget roughly USD 150–300 all in for the journey, excluding the Ambon overnight and the Banda crossing.
Do I need more than one flight?
Yes, at least three legs: Singapore to an Indonesian hub, the hub to Ambon, and Ambon to Banda. There is no shorter version of this route.
Jakarta or Makassar?
Makassar if you want the shortest transit and your dates have slack. Jakarta if your dates are fixed — it is a bigger hub with more onward Ambon departures to fall back on.
Do I need a visa?
It depends on your nationality, and it is checked at the first Indonesian airport rather than on arrival at Ambon. Confirm the current position before you book, since these rules change.
What does the whole trip cost?
Indicatively USD 150–300 one way for the flights, plus USD 10–20 for a sea crossing or IDR 500,000–900,000 for the charter flight, plus an Ambon room.
